What is a DNA paternity
test, and how is it performed?
A DNA paternity test uses DNA samples from the alleged father, child,
and mother to create their genetic profiles—snapshots of each
person’s genetic makeup. Since children inherit their DNA
from their biological mother and father, the child’s genetic
profile should be composed of equal portions from the genetic profiles
of the mother and father.
In a DNA test, buccal swabs are used to collect DNA
from loose cheek cells. DNA is extracted from these cells and purified
for the Polymerase Chain Reaction (PCR). PCR targets specific segments
in the DNA (called markers), amplifying them to detectable levels.
The markers are recorded as a genetic profile. Statistical calculations
are used to determine the probability of paternity based on how
closely the child’s profile matches those of the parents.
